I want to use my Denon Mc6000 MK2 to DJ on ch 2-3 and 4 for Youtube and other internet audio.
Does any of this have to do with virtualDJ and Can anyone help with the configuration?

No not the audio config. That will be the same as if you for instance was recording locally
The MC6000mk2 won't "know" what's on the other end of it
But you need to video broadcast/stream to Youtube
You can do that directly from VDJ using the broadcasting feature, or you can use a third party tool if you prefer

I think he wants to PLAY audio from YouTube.
From YouTube's Terms & Conditions:-
