Just wondering if I can save a loop with a specific stem, i.e. can I save a loop knocking out the vocals then when I release the loop the vocals will turn back on?

You can do that only with action POIs
So, set up an action poi on the start of the loop and use something like this:
loop ? nothing : stem "vocal" 0% & loop 4
Then move a few ms AFTER the loop out point and set up a second POI and use this script
stem "vocal" 50%
