Yes it will.
The only problem you may find is getting hold of Vista 64bit drivers for certain soundcards, so make sure you do some research before buying a soundcard.

I have a Maya 44 sound card and HD video card. Would I have problems with my videos playing?

There are 64bit drivers for the Maya44, so that should be fine.

I'm using a ATI DirertX10 video card. Some video will not play right. I'm running Vista.

Have you tried installing the latest DirectX 9 package. VirtualDJ is design around DirectX 9.
My Nvidia card is a DirectX 10 card, but is backward capable, so running DirectX 9 along side 10 is fine. I don't know whether ATI cards are backward capable but you could always try it.
