I normally DJ with the Numark DDS80, and a pioneer DJM-3000, but I am interested in trying it on a laptop. I purchased the VirtualDJ Pro, and I am thinking about purchasing the Dee Jay Trim 4 & 6 sound card. My question is, can I DJ off the laptop, and send the signal via USB to the Dee Jay Trim and then directly to my amps, or do I have to go through my mixer? Any help will be appreciated.

You can connect the Trim directly to the amp if you want, but it makes more sense to connect it to the mixer. This way you can use the mixer for what it is intended for - mixing :)

Can't you use the mixer in the virtual DJ instead?

you can but that means mixing with the mouse
or use a controller
if you just want to use the onboard mixer in the software and output to
your home stereo or home theater , you can do that ......

Yes you can use the mixer in VDJ, but as a seasoned DJ you will find it combursome mixing with a keyboard/mouse rather than a crossfader.
Now seeing as you have all this kit already, there is no need to invest in a controller for VDJ. Simply connect the trim to the mixer so you have one deck per channel and use the crossfader on the mixer.
You will also be able to cue and PFL on the mixer as well as use the mic input bringing it all together nicely.
Moving to a laptop shouldn't require you to learn a whole new set of skills, you just need to adapt your setup to incorporate the lappy.
Cheers,
Roy

So If I input to my mixer, do I still need the external sound card, or can I split the signal coming from my headphone jack? What I am really looking for is a backup system in case my controller (Numark DDS-80) or my mixer goes out. I am always worrying about something tearing up while I am at a gig...

you should use the soundcard and send the soundcard to the mixer
it's smart to be concerned about problem scenarios and preparation
you should back up your music and/or video to external hard drive (s)
to have backup for the operating system , you need to have backup
laptop or desktop systems ......

Id say with controllers being so SMALL and cheap there really no need to just try and play off your laptop...............just go buy a cheap controller and then do what you do, rock crowds.
Just my thoughts

To re-enforce the "use your mixer" comment, you'll find that if you play in a venue, you'll more often than not find that the connection will be XLR rather than RCA into the amps, so using your mixer will get over this obvious hurdle.
As my 4 deck setup doesn't use a mixer, I built a 4 channel ground loop isolator that has 4 RCA inputs, 4 RCA outputs and two XLR outputs, for those times when I can't be bothered dragging my CDJ's and Mixer along.
